%0 Journal Article %T 星形胶质细胞缺氧缺糖后PKC PKA PKG CaMKⅡ表达的变化及对AQP4的影响 %T Changes of PKC, PKA, PKG and CaMKⅡ expressions in astrocytes after oxygen and glucose deprivation and their effects on AQP4 %A 范秉林,韦俊杰,李燕华,陈 志,韦 旋,李吕力 %A FAN,Bing lin %A WEI,Jun jie %A LI,Yan hua %A et al. %J 中国临床新医学 %J CHINESE JOURNAL OF NEW CLINICAL MEDICINE %@ 1674-3806 %V 12 %N 12 %D 2019 %P 1276-1279 %K 星形胶质细胞;缺氧;缺糖 %K Astrocytes;Oxygen deprivation;Glucose deprivation %X [摘要] 目的 探讨星形胶质细胞在缺氧、缺糖情况下蛋白激酶C(PKC)、蛋白激酶A(PKA)、蛋白激酶G(PKG)及钙/钙调素依赖性蛋白激酶Ⅱ(CaMKⅡ)的表达变化情况。方法 取2 d龄新生SD大鼠的大脑皮层进行星形胶质细胞的纯化与培养,通过缺氧、缺糖处理建立缺血细胞模型。以实验处理方式的不同分为对照组和模型组,通过RT-PCR法检测星形胶质细胞水通道蛋白4(AQP4)表达的变化,采用Western blot法检测星形胶质细胞PKC、PKA、PKG和CaMKⅡ的表达情况。比较两组各项指标的差异。结果 模型组的AQP4表达水平显著高于对照组,差异有统计学意义(P<0.05)。模型组的PKC表达水平低于对照组,CaMKⅡ表达水平高于对照组,差异有统计学意义(P<0.05)。PKA、PKG的表达水平在两组间比较差异无统计学意义(P>0.05)。结论 缺氧、缺糖导致的星形胶质细胞的PKC表达水平下降及CaMKⅡ表达水平升高,可能与星形胶质细胞水肿有关。 %X [Abstract] Objective To explore the expression changes of protein kinase C(PKC), protein kinase A(PKA), protein kinase G(PKG) and calcium/calmodulin-dependent protein kinase Ⅱ(CaMKⅡ) in cerebral astrocytes under oxygen and glucose deprivation. Methods The cerebral cortex of 2-day-old newborn SD rats was purified and cultured for astrocytes, and the ischemic cell model was established by oxygen and glucose deprivation. The control group and the model group were grouped according to different experimental methods. Reverse transcription-polymerase chain reaction(RT-PCR) was used to detect the expressions of aquaporin-4(AQP4), and Western blot was used to detect the expressions of PKC, PKA, PKG and CaMKⅡ in astrocytes. The differences in the indicators were compared between the two groups. Results The expression of AQP4 in the model group was significantly higher than that in the control group(P<0.05). The expression of PKC in the model group was significantly lower than that in the control group(P<0.05). The expression of CaMKⅡ in the model group was significantly higher than that in the control group(P<0.05). The expressions of PKA and PKG were not significantly different between the two groups(P>0.05).
