基于芯片数据库Oncomine分析CTGF基因在多发性骨髓瘤中的表达和临床意义

[摘要] 目的 研究结缔组织生长因子(connective tissue growth factor,CTGF)基因在多发性骨髓瘤(multiple myeloma,MM)及正常组织中的表达和临床意义。方法 检索Oncomine数据库中所有关于CTGF的数据集,对其在MM中的作用进行Meta分析。结果 Oncomine数据库中共收集了456项不同肿瘤中关于CTGF的研究,与正常组织相比,其表达差异具有统计学意义的共52项,其中CTGF表达增高28项,表达降低24项。共4项研究中的8个数据集包含379例样本涉及CTGF在MM组织与正常组织中的表达,与正常组织相比,5个数据集中CTGF在癌组织中呈显著高表达(P<0.05),2个数据集中其表达差异无统计学意义(P>0.05),1个数据集中其统计值P=1.000。Meta分析结果显示,CTGF在癌组织与正常组织中的表达差异无统计学意义(P>0.05);进一步分析发现,GTGF表达量与预后无显著关联性(P>0.05)。结论 CTGF在正常组织和MM组织中表达无明显差异,不能作为MM的一个潜在的标志物,不能对预后进行预测。

[Abstract] Objective To study the expressions of connective tissue growth factor(CTGF) gene in multiple myeloma(MM) and normal tissues and their clinical significances. Methods Datasets about CTGF in Oncomine database were retrieved and the role of CTGF in MM was analyzed by Meta analysis. Results A total of 456 studies on the expressions of CTGF in different tumors were collected in the Oncomine database. Compared with those in the normal tissues, the CTGF expressions were significantly different in 52 studies among which the CTGF expressions were higher in 28 studies and lower in 24 studies. Eight datasets in the four studies included 379 samples involving the expressions of CTGF in the MM and normal tissues. Compared with those in the normal tissues, the expressions of CTGF in the cancer tissues were significantly higher in 5 datasets(P<0.05), but there were no significant differences in 2 datasets(P>0.05), in which the statistic value of CTGF was P=1.000 in 1 dataset. Meta analysis showed that there was no significant difference in the expression of CTGF between the cancer and normal tissues(P>0.05); Further analysis showed that there was no correlation between CTGF expression and prognosis(P>0.05). Conclusion There is no significant difference in CTGF expression between normal and MM tissues. CTGF can not be used as a potential marker of MM to predict the prognosis of MM.
