DOK3 PTK7在不同级别结肠腺瘤组织中的表达及意义探讨

摘要:
[摘要] 目的 分析对接蛋白3(DOK3)和蛋白酪氨酸激酶7(PTK7)在不同级别结肠腺瘤组织中的表达情况,探讨其在结肠癌的发生、发展机制中的作用。方法 收集2015-01~2018-12广西壮族自治区南溪山医院经病理科确诊的结肠腺瘤伴低级别异型增生(LGD)标本22例,结肠腺瘤伴高级别异型增生(HGD)标本23例,结肠癌标本22例,瘤旁非肿瘤黏膜组织标本20例。采用免疫组织化学染色法检测不同类型标本DOK3和PTK7的表达情况。结果 DOK3和PTK7均主要表达于腺上皮的胞浆和胞膜。瘤旁非肿瘤黏膜和LGD组织的DOK3高表达率均显著高于HGD和结肠癌组织(P<0.05);HGD组织的DOK3高表达率也显著高于结肠癌组织(P<0.05)。结肠癌和HGD组织的PTK7高表达率均显著高于瘤旁非肿瘤黏膜和LGD组织(P<0.05);结肠癌组织的PTK7高表达率也显著高于HGD组织(P<0.05)。结论 DOK3可能是作为抑癌因子,而PTK7作为促癌因子参与结肠癌的发生、发展。

Expressions and significances of DOK3 and PTK7 in different grades of colon adenoma tissues

Abstract:
[Abstract] Objective To analyze the expressions of docking protein 3(DOK3) and protein tyrosine kinase 7(PTK7) in different grades of colon adenoma tissues, and to explore their roles in the occurrence and development of colon cancer. Methods The specimens from 22 cases of colon adenoma with low-grade dysplasia(LGD), 23 cases of colon adenoma with high-grade dysplasia(HGD), 22 cases of colon cancer and 20 cases of non-tumor mucosal tissue adjacent to the tumor who were diagnosed by the Department of Pathology, Nanxishan Hospital of Guangxi Zhuang Autonomous Region were collected during January 2015 and December 2018. Immunohistochemical staining method was used to detect the expressions of DOK3 and PTK7 in different types of specimens. Results Both DOK3 and PTK7 were mainly expressed in the cytoplasm and membrane of glandular epithelium. The high expression rates of DOK3 in non-tumor mucosal tissues adjacent to the tumor and LGD tissues were significantly higher than those in HGD tissues and colon cancer tissues(P<0.05). The high expression rate of DOK3 in HGD tissues was also significantly higher than that in colon cancer tissues(P<0.05). The high expression rates of PTK7 in colon cancer tissues and HGD tissues were significantly higher than those in non-tumor mucosal tissues adjacent to the tumor and LGD tissues(P<0.05). The high expression rate of PTK7 in colon cancer tissues was also significantly higher than that in HGD tissues(P<0.05). Conclusion DOK3 may act as a tumor suppressor, and PTK7 may act as a cancer promoter involving in the occurrence and development of colon cancer.
