犬下颌牵张成骨不同时期六种染色方法的应用效果比较

[摘要] 目的 比较六种染色方法在犬下颌牵张成骨早期、中期、后期的染色效果,为研究者选择合适的染色方法提供参考。方法 选用1岁雄性比格犬,经麻醉后手术造模,分别在牵张成骨早期、中期、后期行安乐死后取牵张成骨区组织,脱钙切片后分别采用HE染色、Masson染色、亚甲基蓝-酸性品红染色、Von Kossa染色、改良茜素红S染色、改良番红O-固绿染色对切片进行染色,分析比较染色效果。结果 HE染色可观察牵张成骨区基础结构。Masson染色颜色对比鲜明,结构清晰,可以清晰地区分胶原纤维与肌纤维,以及成熟骨质与新生骨质。改良番红O-固绿染色与亚甲基蓝-酸性品红染色在牵张成骨早期、中期的区分度不佳,但在牵张成骨后期可清晰区分新生骨组织与成熟骨组织。Von Kossa染色和改良茜素红S染色因标本已脱钙而效果不佳。结论 对于牵张成骨区域的染色研究,Masson染色法在牵张成骨早期、中期、后期均有较好的染色效果。改良番红O-固绿染色和亚甲基蓝-酸性品红染色仅适用于牵张成骨后期。Von Kossa染色法和改良茜素红S染色法不适用于脱钙后的组织切片染色。

Comparison of application effects of six staining methods at different stages of mandibular distraction osteogenesis in beagles

Abstract:
[Abstract] Objective To compare the staining effects of six staining methods at the early, middle and late stages of mandibular distraction osteogenesis in beagles, so as to provide reference for researchers to select the appropriate staining methods. Methods One-year-old male beagles were selected for surgical modeling after anesthesia. The tissues of the distraction osteogenesis area were taken respectively at the early, middle and late stages of distraction osteogenesis in beagles after euthanasia. After decalcification, the sections were stained by hematoxylin and eosin(HE) staining, Masson staining, methylene blue-acidic magenta staining, Von Kossa staining and modified alizarin red S staining, and modified safranin-O fast green staining, respectively, and the effects of staining were compared. Results HE staining could be used to observe the basic structure of the distraction osteogenesis area. The Masson staining showed sharp color contrast and clear structure, which could clearly distinguish collagen fibers from muscle fibers and mature bones from new bones. The modified safranin-O fast green staining and the methylene blue-acidic magenta staining were poorly differentiated at the early and middle stages of the distraction osteogenesis, but the two staining methods could clearly distinguished mature bones from new bones at the late stage of distraction osteogenesis. The Von Kossa staining and the modified alizarin red S staining were not very effective because the samples had been decalcified. Conclusion For the staining study of the distraction osteogenesis area, Masson staining has better staining effects at the early, middle and late stages of distraction osteogenesis. The modified safranin-O fast green staining and the methylene blue-acidic magenta staining are only suitable for the late stage of distraction osteogenesis. The Von Kossa staining method and the modified alizarin red S staining method are not suitable for the staining of tissue sections after decalcification.
