miR-630对肾透明细胞癌发生发展的影响及机制研究

摘要:
[摘要] 目的 探究miR-630对肾透明细胞癌(ccRCC)的发生发展的影响及其机制。方法 选择2020年7月至2021年7月河北工程大学附属医院收治的ccRCC患者86例,术中取肿瘤组织和癌旁正常组织(距癌灶边缘>5 cm),通过实时荧光定量聚合酶链式反应(RT-qPCR)法检测肿瘤组织和癌旁正常组织中miR-630表达水平,分析其与ccRCC患者临床病理特征的关联性。分析ccRCC组织中miR-630表达水平与凋亡标志蛋白表达水平的相关性。选择786-O细胞进行实验,设计miR-630 NC组、miR-630 mimic组和miR-630 inhibitor组(分别为miR-630的对照、过表达和抑制组),分析miR-630表达对细胞增殖活力、凋亡率以及凋亡标志蛋白表达水平的影响。结果 ccRCC组织中miR-630水平显著高于癌旁正常组织(P<0.05)。高水平的miR-630与更大的肿瘤直径、高临床分期及淋巴结转移有关(P<0.05)。与癌旁正常组织相比,ccRCC组织中caspase-3蛋白表达水平显著降低(P<0.05),B淋巴细胞瘤-2基因(Bcl2)蛋白水平显著升高(P<0.05)。ccRCC组织中miR-630表达水平与caspase-3蛋白表达水平呈负相关(r=-0.489,P=0.011),与Bcl2蛋白表达水平呈正相关(r=0.516,P=0.004)。过表达miR-630可显著促进细胞增殖活力,抑制细胞凋亡率以及caspase-3蛋白表达水平,促进Bcl2蛋白表达水平(P<0.05)。降低miR-630表达可显著抑制细胞增殖活力,促进细胞凋亡及caspase-3蛋白表达水平,抑制Bcl2蛋白表达水平(P<0.05)。结论 miR-630在ccRCC组织中表达上调,高水平的miR-630可抑制细胞凋亡,并与ccRCC患者的高临床分期和淋巴结转移情况有关。

Abstract:
[Abstract] Objective To explore the effects of micro ribonucleic acid-630(miR-630) on the occurrence and development of clear cell renal cell carcinoma(ccRCC) and their mechanisms. Methods Eighty-six patients with ccRCC who were admitted to Affiliated Hospital of Hebei University of Engineering from July 2020 to July 2021 were selected. The tumor tissues and the adjacent normal tissues(more than 5 cm away from the tumor focus) were taken during the operation, and the expression levels of miR-630 in the tumor tissues and the adjacent normal tissues were detected by using real-time quantitative polymerase chain reaction(RT-qPCR). The correlations of the expression levels of miR-630 in the tumor tissues and the adjacent normal tissues with the clinicopathological features of the ccRCC patients were analyzed. The correlation between the expression level of miR-630 and the expression level of apoptosis marker protein in the ccRCC tissues was analyzed. The 786-O cells were selected for experiment, and miR-630 NC group, miR-630 mimic group and miR-630 inhibitor group(They were the control, overexpression and inhibition groups of miR-630, respectively) were designed. The effects of miR-630 expression on cell proliferation activity, apoptosis rate and the expression level of apoptosis marker protein were analyzed. Results The level of miR-630 in the ccRCC tissues was significantly higher than that in the adjacent normal tissues(P<0.05). High levels of miR-630 were associated with larger tumor diameter, high clinical stage and lymphatic metastasis(P<0.05). Compared with those in the adjacent normal tissues, the expression levels of caspase-3 protein in the ccRCC tissues were significantly decreased(P<0.05), and the levels of B cell lymphoma leukemia-2(Bcl2) protein in the ccRCC tissues were significantly increased(P<0.05). The expression level of miR-630 in the ccRCC tissues was negatively correlated with the expression level of caspase-3 protein(r=-0.489, P=0.011), and positively correlated with the expression level of Bcl2 protein(r=0.516, P=0.004). The overexpression of miR-630 could significantly promoted cell proliferation activity, inhibited cell apoptosis rate and the expression level of caspase-3 protein, and promoted the expression level of Bcl2 protein(P<0.05). Reducing the expression of miR-630 could significantly inhibit cell proliferation activity, promoted cell apoptosis and the expression level of caspase-3 protein, and inhibited the expression level of Bcl2 protein(P<0.05). Conclusion The level of miR-630 is up-regulated in ccRCC tissues, and high level of miR-630 can inhibit cell apoptosis, and is associated with high clinical stage and lymphatic metastasis in ccRCC patients.
