熊去氧胆酸抑制人肝星状细胞活化和NIX蛋白表达的实验研究

摘要:
[摘要] 目的 研究NIX蛋白的表达变化与熊去氧胆酸(UDCA)抑制人肝星状细胞(HSC)活化作用的关系。方法 人肝星状细胞-LX2细胞实验分为四组:溶媒对照组、UDCA(0.5 μM)组、TGF-β1(5 ng/ml)组、TGF-β1(5 ng/ml)+UDCA(0.5 μM)组。通过Western blot检测比较各组间α-SMA、NIX的表达差异。结果 TGF-β1刺激LX2细胞后,α-SMA、NIX表达分别较对照组增加60.9%、51.0%;UDCA抑制TGF-β1活化LX2细胞后,α-SMA的表达抑制率达55.6%,与其显著下调NIX的表达呈正相关。结论 UDCA下调HSC细胞NIX蛋白的表达,可能为UDCA抗肝纤维化的药理新机制。

An in vitro research of ursodesoxycholic acid inhibiting the activation of hepatic stellate cell and the expression of NIX protein

Abstract:
[Abstract] Objective To study the relationship between the expression changes of NIP3-like protein X(NIX) in hepatic stellate cell(HSC) and the inhibitory role of ursodesoxycholic acid(UDCA) on activation of HSC.Methods HSC-LX2 cells were divided into four groups: vehicle control group,UDCA(0.5 μM) group, TGF-β1(5 ng/ml) group, and TGF-β1(5 ng/ml)+UDCA(0.5 μM) group. The expressions of α-SMA and NIX were detected by Western blot and were compared among the different groups.Results After HSC-LX2 cells were stimulated by TGF-β1, the expression of α-SMA in LX2 cells was 60.9% higher than that in the control group, and the expression of NIX was simultaneously elevated with an increase of 51.0%. The expression of a-SMA was inhibited 55.6% by UDCA in LX2 cells which were activated by TGF-β1, and that was positively correlated with the significantly down-regulated expression of NIX.Conclusion UDCA reduces the expression of NIX protein in HSC, which might be one of the pharmacological mechanisms of UDCA to counteract liver fibrosis.
