脑胶质瘤中MAGE-D4和CORO1C mRNA表达水平及其临床意义

[摘要] 目的 分析脑胶质瘤中黑色素瘤相关抗原(MAGE)-D4和肌动蛋白结合蛋白CORO1C mRNA的表达水平,探讨二者的相关性及临床意义。方法 通过基因表达谱交互式分析(GEPIA)数据库收集脑胶质瘤样本数据681例[低级别脑胶质瘤(LGG)518例,胶质母细胞瘤(GBM)163例],正常脑组织样本数据207例,比较其MAGE-D4和CORO1C mRNA的表达水平。进一步收集62例脑胶质瘤组织和11例正常脑组织临床样本,应用逆转录-定量聚合酶链反应(RT-qPCR)检测其MAGE-D4和CORO1C mRNA表达水平并进行比较,分析MAGE-D4和CORO1C mRNA表达水平与脑胶质瘤患者临床指标的关联性。应用小干扰RNA(siRNA)技术沉默脑胶质瘤细胞株MAGE-D4的表达,检测CORO1C mRNA的表达变化情况。结果 GEPIA数据库数据分析结果显示,GBM和LGG组织的MAGE-D4和CORO1C mRNA表达水平均高于正常脑组织,差异有统计学意义(P<0.05)。针对临床样本的RT-qPCR检测结果也显示,脑胶质瘤组织中MAGE-D4和CORO1C mRNA的表达水平显著高于正常脑组织(P<0.05)。MAGE-D4 及CORO1C mRNA的表达水平与患者年龄、性别、世界卫生组织(WHO)分级、病理类型、卡氏评分(KPS)、肿瘤最长径,以及Ki-67、胶质纤维酸性蛋白(GFAP)、S-100和波形蛋白(Vimentin)的表达水平均无显著关联(P>0.05)。Pearson相关分析结果显示,临床脑胶质瘤样本组织中MAGE-D4与CORO1C mRNA表达水平呈正相关(r=0.848,P=0.000)。细胞实验结果显示,经MAGE-D4 siRNA沉默后,U87 MG和U251细胞CORO1C mRNA的表达水平显著下降(P<0.05)。结论 MAGE-D4和CORO1C mRNA在脑胶质瘤组织中高表达,且二者表达水平呈正相关。MAGE-D4和CORO1C可能协同影响脑胶质瘤的发生、发展过程,其具有成为脑胶质瘤分子靶向治疗靶点的潜力。

Expression levels of MAGE-D4 and CORO1C mRNA in glioma and their clinical significances

Abstract:
[Abstract] Objective To analyze the expression levels of melanoma-associated antigen family member D4(MAGE-D4) and actin-binding protein CORO1C mRNA in glioma, and to explore their correlation and clinical significances. Methods The data of six hundred and eighty-one glioma samples[518 cases of low-grade glioma(LGG) and 163 cases of glioblastoma(GBM)] and the data of 207 cases of normal brain tissue samples were collected through Gene Expression Profiling Interactive Analysis(GEPIA) database, and the expression levels of MAGE-D4 and CORO1C mRNA were compared among them. The clinical samples of 62 cases of glioma tissues and 11 cases of normal brain tissues were further collected, and reverse transcription-quantitative polymerase chain reaction(RT-qPCR) was used to detect the expression levels of MAGE-D4 and CORO1C mRNA, and the expression levels of MAGE-D4 and CORO1C mRNA were compared between the two clinical samples. The correlation between the expression levels of MAGE-D4 and CORO1C mRNA and the clinical indicators of the glioma patients was analyzed. Small interfering RNA(siRNA) technology was used to silence the expression of glioma cell line MAGE-D4, and the changes in the expression of CORO1C mRNA was detected. Results The results of GEPIA database data analysis showed that the expression levels of MAGE-D4 and CORO1C mRNA in the GBM and LGG tissues were higher than those in the normal brain tissues, and the differences were statistically significant(P<0.05). The RT-qPCR detection results of the clinical samples also showed that the expression levels of MAGE-D4 and CORO1C mRNA in the glioma tissues were significantly higher than those in the normal brain tissues(P<0.05). The expression levels of MAGE-D4 and CORO1C mRNA were not significantly correlated with the patient′s age, gender, World Health Organization(WHO) grade, pathological type, Karnofsky performance score(KPS), the longest diameter of the tumor, and the expression levels of Ki-67, glial fibrillary acidic protein(GFAP), S-100 and Vimentin(P>0.05). The results of Pearson correlation analysis showed that there was a positive correlation between the expression levels of MAGE-D4 and CORO1C mRNA in the clinical glioma sample tissues(r=0. 848, P=0.000). The results of cell experiment showed that the expression levels of CORO1C mRNA in U87 MG and U251 cells decreased significantly after the U87 MG and U251 cells were silenced by MAGE-D4 siRNA(P<0.05). Conclusion MAGE-D4 and CORO1C mRNA are highly expressed in glioma tissues, and their expression levels are positively correlated. MAGE-D4 and CORO1C may synergistically affect the occurrence and development of glioma, and they have the potential to become targets for molecular targeted therapy of glioma.
