阿托伐他汀对慢性子宫内膜炎大鼠的治疗作用及其机制研究

摘要:
[摘要] 目的 探讨阿托伐他汀对慢性子宫内膜炎(CE)大鼠的治疗作用及相关机制。方法 选择无特定病原体(SPF)级雌性SD大鼠32只,随机将其分为假手术组、模型组、阿托伐他汀低剂量组(10 mg/kg)和阿托伐他汀高剂量组(20 mg/kg),每组8只。采用子宫内注射苯酚胶浆剂建立CE大鼠模型。连续灌胃给药干预14 d后取大鼠子宫组织,经HE染色观察大鼠子宫组织病理学改变。应用实时荧光定量聚合酶链式反应(RT-qPCR)法检测大鼠子宫组织白细胞介素-1β(IL-1β)和基质金属蛋白酶-9(MMP-9)mRNA的表达水平。应用免疫组化法检测大鼠子宫组织IL-1β和MMP-9蛋白的表达水平。结果 与模型组比较,阿托伐他汀高剂量组大鼠子宫内膜上皮细胞增生、变性坏死、炎细胞浸润情况得到显著改善。阿托伐他汀低剂量组、阿托伐他汀高剂量组大鼠子宫组织IL-1β和MMP-9 mRNA表达水平和蛋白阳性表达率低于模型组,差异有统计学意义(P<0.05)。结论 阿托伐他汀可以改善CE大鼠的炎症反应,其机制可能与下调IL-1β和MMP-9的表达有关。

A study on the therapeutic effect of atorvastatin on chronic endometritis in rats and its mechanisms

Abstract:
[Abstract] Objective To investigate the therapeutic effect of atorvastatin on chronic endometritis(CE) in rats and its related mechanisms. Methods Thirty-two specific pathogen free(SPF) female Sprague-Dawley(SD) rats were selected and randomly divided into sham operation group, model group, low-dose atorvastatin group(10 mg/kg) and high-dose atorvastatin group(20 mg/kg), with 8 rats in each group. The CE rat model was established by intrauterine injection of phenol mucilage. After 14 days of continuous intragastric administration, the rat uterine tissues were collected, and the histopathological changes of the rat uteruses were observed by hematoxylin and eosin(HE) staining. The expression levels of interleukin-1β(IL-1β) and matrix metalloproteinase-9(MMP-9) mRNA in the rat uterine tissues were detected by real-time fluorescence quantitative polymerase chain reaction(RT-qPCR). The expression levels of IL-1β and MMP-9 in the rat uterine tissues were detected by immunohistochemical method. Results Compared with those in the model group, the endometrial epithelial cell proliferation, degeneration and necrosis, and inflammatory cell infiltration of the rats in the high-dose atorvastatin group were significantly improved. The mRNA levels and protein positive expression rates of IL-1β and MMP-9 in the rat uterine tissues in the low-dose atorvastatin group and high-dose atorvastatin group were lower than those in the model group, and the differences were statistically significant(P<0.05). Conclusion Atorvastatin can improve the inflammatory response in the CE rats and its mechanisms may be related to down-regulating the expressions of IL-1β and MMP-9.
