目的 本研究旨在探讨IL-6在HIV合并TM感染的人群中的表达和作用。方法 在南宁市第四人民医院招募HIV合并TM感染的病人和HIV不合并TM感染的未进行抗病毒治疗的病人,收集病人的电子病历信息,采集两个人群的外周血,分离PBMC,通过RT-PCR检测PBMC中IL-6基因表达和通过流式细胞术检测PBMC中巨噬细胞的IL-6蛋白表达水平。通过t检验、卡方检验比较HIV合并TM感染组和不合并TM感染组之间的人口学资料,用t检验对两组人群mRNA、蛋白水平的比较进行分析。结果 本研究共纳入了27例艾滋病病人,其中合并TM感染的病人12例,平均年龄为40.83岁,不合并TM感染的病人15例,平均年龄为52.89岁。性别两组均以男性为主,CD4+T细胞计数在TM感染组全部低于100cells/μL,而不合并TM感染的病人60%低于100cells/μL。IL-6的mRNA表达水平在合并TM感染组高于未合并TM感染组,但差异无统计学意义( t=1.014,p=0.327)。流式细胞术结果显示IL-6在HIV合并TM感染的人群表达高于HIV不合并TM感染的人群表达,差异有统计学意义(t=2.640,P=0.039)。结论 HIV合并TM感染人群IL-6高表达,导致巨噬细胞免疫过度激活,可能在HIV合并TM感染促进艾滋病病人死亡可能起关键作用。 |

Objective To explore the expression and its role of IL-6 in HIV/TM co-infection patients. Methods Recruited ART-na?ve ,HIV patients with and without TM infection in the Fourth People's Hospital of Nanning, collected peripheral blood and isolated peripheral blood mononuclear cells(PBMC). Detect the gene expression of IL-6 in PBMC by RT-PCR, and the protein expression in macrophage by Flow cytometry. Chi-square test and t-test were used to analyze the demographic data of the combined TM infection group and the non-merger TM infection group. t-test was used to analyze the mRNA and protein levels of the two groups. Results This study recruited 12 AIDS patients with TM infection, mean aged 40.83 years old,and 15 AIDS patients without TM infection, mean aged 52.89 years old. The most of patients were male. The CD4+T cell counts of all HIV/TM co-infection patients were less than 100 cells/μL, the 60% were less than 100 cells/μL in the non-merger TM infection group. The mRNA expression of IL-6 of the HIV/TM co-infection group higher than the non-merger TM infection group ,but had no significant (t=1.014,p=0.327), while protein expression of IL-6 had a significant tendency to increase in the HIV/TM co-infected patients (t=2.640,P=0.039). Conclusion The high IL-6 expression the combined TM infection group lead to immune hyperactivation in macrophages, and may play a vital role to the death of HIV patients combined TM infection. |
