TM5275对TGF-β1诱导的人系膜细胞增殖、凋亡和细胞外基质的影响

摘要:
目的 探讨TM5275对TGF-β1诱导的人肾小球系膜细胞增殖、凋亡和细胞外基质的影响。方法 体外培养人肾小球系膜细胞,实验分为对照组,TGF-β1组,TGF-β1+TM5275干预组,培养24h~48h。MTT法检测系膜细胞的增殖,Annexin Ⅴ/PI双染色法检测细胞凋亡。采用Western blot法检测PAI-1、a-SMA、Fibronectin、Collagen-IV、Bax、Bcl-2和caspase-3。结果TM5275明显抑制TGF-β1诱导的系膜细胞增殖、并促进其凋亡(P<0.05); TGF-β1诱导系膜细胞的PAI-1和a-SMA表达显著增多及细胞外基质Fibronectin、Collagen-IV的表达显著增加(P<0.05)可被TM5275显著抑制(P<0.05)。TM5275干预后,促凋亡蛋白Bax、caspase-3表达明显增加(P<0.05),阻止凋亡蛋白Bcl-2未见明显表达。结论TM5275可抑制TGF-β1诱导的系膜细胞增殖和细胞外基质的表达,同时通过诱导促凋亡蛋白的表达,促进异常增殖的系膜细胞凋亡。其具有治疗系膜增生性肾小球肾炎的潜在前景。

Abstract:
Objective To study the effects of TM5275 on the proliferation apoptosis and extracellular matrix of human mesangial cells induced by TGF-β1.Methods Human mesangial cells were cultured in vitro.The experiment was divided into the control group,The TGF-β1 group and The TM5275 intervention group (TGF-β1+TM5275). The cells were cultured for 24 to 48 hours. MTT assay was used to detect the proliferation of mesangial cells and Annexin V/PI double staining was used to detect apoptosis. PAI-1, a-SMA, Fibronectin, Collagen-IV, Bax, Bcl-2 and caspase-3 were detected by Western blot.Results TM5275 significantly inhibited the proliferation of mesangial cells induced by TGF-β1 and promoted mesangial cells apoptosis(P<0.05).The expression increased of PAI-1 ,a-SMA ,extracellular matrix Fibronectin and Collagen-IV in mesangial cells induced by TGF-β1 was significantly inhibited by TM5275(P<0.05). After TM5275 intervention, the expression of pro-apoptotic proteins Bax and caspase-3 significantly increased(P<0.05),while the expression of anti-apoptotic protein Bcl-2 did not increase.Conclusion TM5275 can inhibit the proliferation of mesangial cells and the expression of extracellular matrix induced by TGF-β1.At the same time,TM5275 can promote the apoptosis of abnormal proliferation mesangial cells by inducing the expression of pro-apoptotic proteins.It has potential prospects for the treatment of mesangial proliferative glomerulonephritis.
