目的 观察EMX2和β-catenin、Cyclin D1在正常结直肠黏膜、结直肠癌组织中的表达,探讨结直肠癌发展过程中EMX2和β-catenin、Cyclin D1的关系。方法 S-P法检测EMX2和β-catenin、Cyclin D1在正常结直肠黏膜、结直肠癌组织中蛋白表达,并分析EMX2和β-catenin、Cyclin D1与结直肠癌临床病理特征之间及患者预后的关系。结果 1 在正常结直肠黏膜、结直肠癌组织中EMX2阳性表达率分别为76.7%和38.2%(P<0.001),EMX2阳性表达率与结直肠癌临床分期及淋巴结转移有关。在正常结直肠黏膜、结直肠癌组中β-catenin阳性表达率分别为6.7%、65.5%;Cyclin D1阳性表达率分别为10.0%、63.6%。β-catenin与Cyclin D1阳性表达率与结直肠癌的临床分期、肿瘤分化程度、淋巴结是否转移相关(P<0.05)。2 在结直肠癌组织中EMX2和β-catenin呈负一致性 (kappa值=-0.389,P=0.000); EMX2 与Cyclin D1表达不相关(kappa值=-0.161, P=0.054)。3 在结直肠癌中应用Kaplan-Meier 生存分析,表明结直肠癌组织中EMX2阳性表达与患者较好的预后相关(P<0.032)。结论 结直肠癌组织中EMX2阳性表达减少,EMX2低表达与结直肠癌患者的临床分期、淋巴结转移及预后密切关系。另外在结直肠癌患者中检测EMX2,可能成为判断患者临床分期与预后的一个指标。 |

Objective To investigate the expression levels of empty spiracles homeobox 2 (EMX2) and β-catenin、cyclin D1 in colorectal cancers and their relationships between the occurrence, development, and prognosis of colorectal cancer. Methods The immunohistochemical S-P method was used to detect the expressions of the EMX2 and β-catenin、cyclin D1 proteins in colorectal tissues .The correlations of EMX2 and β-catenin、cyclin D1 with clinicopathologic features were analyzed in colorectal cancers. Results The positive rates of EMX2 expression was 76.7% in normal colorectal tissues and in colorectal cancer tissues the expression was 38.2%. The expression rates of the EMX2 were significantly different between the colorectal normal tissues and colorectal cancers (P<0.001). The expression level of the EMX2 in colorectal cancers tissues was associated with clinic Stage, and Grade ,Lymphatic metastasis(P<0.05). Spearman's correlation revealed that EMX2 expression was significantly correlated with β-catenin expression in the 110 colorectal cancers (kappa =-0.389,P=0.000). Kaplan-Meier survival analyses revealed that the decreasing expression of the EMX2 was significantly poorer survival of colorectal cancers patients (P=0.032). Conclusion EMX2 are reduced in colorectal cancers, suggesting that the reduced expression was associated with Stage , Grade ,Lymphatic metastasis .Forthemore the expression of EMX2 was correlated with colorectal cancers prognostic. EMX2 could be used as potential prognostic markers. |
