异硫氰酸苯乙酯联合阿霉素诱导的骨肉瘤细胞凋亡

摘要:
目的 本研究旨在探索异硫氰酸苯乙酯(PEITC)对阿霉素(ADM)诱导的骨肉瘤细胞凋亡的影响。 方法 分别使用异硫氰酸苯乙酯或者阿霉素以及异硫氰酸苯乙酯联合阿霉素处理U2-OS骨肉瘤细胞系细胞24小时,观察它们的剂量抑制作用。 结果 当达到半数抑制浓度(IC50)时异硫氰酸苯乙酯和阿霉素浓度分别为4.37μM和6.61μg/ml。当用异硫氰酸苯乙酯联合阿霉素处理U2-OS细胞系时,抑制作用明显增强,同样达到IC50时异硫氰酸苯乙酯联合阿霉素分别减少到3μM和2μg/ml。TUNEL法表明单独使用异硫氰酸苯乙酯或者阿霉素处理时,U2-OS骨肉瘤细胞系的存活率降低。但是,若联合使用上述两种药物处理U2-OS骨肉瘤细胞系时,其存活率下降更为显著。我们还观察到,在使用异硫氰酸苯乙酯或阿霉素以及异硫氰酸苯乙酯联合阿霉素处理U2-OS骨肉瘤细胞系后,细胞中caspase-3的表达显著增强。 结论 以上结果表明,异硫氰酸苯乙酯增强了阿霉素对骨肉瘤细胞诱导凋亡的作用。

APOPTOSIS OF OSTEOSARCOMA CELLS INDUCED BY PHENYLETHYL ISOTHIOCYANATE COMBINED WITH ADRIAMYCIN

Abstract:
Objective To investigate the effect of phenylethyl isothiocyanate (PEITC) on adriamycin (ADM) induced apoptosis in osteosarcoma cells. Methods PEITC, ADM, PEITC and ADM in combination with doxorubicin were used to treat U2-OS osteosarcoma cell lines for 24 hours, and their dose inhibition was observed. Results The concentrations of PEITC and doxorubicin were 4.37 μM and 6.61 μg/ml, respectively, when the inhibition concentration (IC50) was reached. When the U2-OS cell line was treated with PEITC and ADM, the inhibition was significantly enhanced, and when the same level of IC50 was reached, PEITC and ADM were reduced to 3 μM and 2μg/ml, respectively. TUNEL showed that the survival rate of U2-OS osteosarcoma cell lines was reduced when treated with PEITC or ADM alone. However, when U2-OS cell lines were treated with the two drugs in combination, the survival rate decreased even more significantly. We also observed a significant increase in caspase-3 expression in U2-OS osteosarcoma cell lines treated with PEITC or ADM and PEITC in combination with ADM. Conclusion The above results indicated that PEITC enhanced the apoptotic effect of ADM on osteosarcoma cells.
