DOK3、PTK7在不同级别结肠腺瘤组织中的表达及意义

摘要:
目的:检测DOK3、PTK7在不同级别结肠腺瘤、结肠癌及瘤旁正常组织中的表达,探讨结肠癌前病变的发展机制。方法:分4组:①结肠腺瘤伴低级别异型增生(LGD);②结肠腺瘤伴高级别异型增生(HGD);③结肠癌组;④瘤旁正常组织;用免疫组化分别测DOK3、PTK7的表达。结果:①DOK3在HGD组和结肠癌组的高表达率均低于LGD组、瘤旁正常组织(P <0.05), HGD组高于结肠癌组(P <0.05)。②PTK7在HGD组及结肠癌组的高表达率均高于LGD组、瘤旁正常组织(P <0.05);结肠癌组高于HGD组(P <0.05)。结论:DOK3在结肠腺瘤级别越高时表达量越低,而PTK7在结肠癌中的表达量最高,且在结肠腺瘤级别越高时表达量越高,DOK3可能作为抑癌基因、PTK7作为癌基因参与结肠癌发生、发展。

The expression and significance of DOK3 and PTK7 in different grades of colon adenoma

Abstract:
Objective To detect the expression of DOK3 and PTK7 in different grades of colon adenoma, colon cancer and normal tissues adjacent to the tumor, and to explore the development mechanism of colon precancerous lesions. Mthods The experiment was divided into 4 groups: ① colon adenoma with low-grade dysplasia(LGD); ② colon adenoma with high-grade dysplasia(HGD);③ colon cancer group; ④ normal tissue adjacent to the tumor; The expression of DOK3 and PTK7 were determined by immunohistochemistry. Results ① The high expression rate of DOK3 in the HGD group and colon cancer group was lower than that in the LGD group and normal tissue adjacent to tumor (P <0.05), and the HGD group was higher than the colon cancer group (P <0.05). ② The high expression rate of PTK7 in the HGD group and colon cancer group was higher than that in the LGD group and normal tissues adjacent to the tumor (P <0.05); the colon cancer group was higher than the HGD group (P <0.05). Conclusion The higher the level of colon adenoma, the lower the expression level of DOK3, while the highest expression level of PTK7 in colon cancer, and the higher the level of expression at higher levels of colon adenoma. DOK3 may act as a tumor suppressor gene and PTK7 as a tumor suppressor gene, Oncogenes are involved in the occurrence and development of colon cancer.
