| 甲氨蝶呤（methotrexate，MTX）是二氢叶酸还原酶抑制剂，临床上常用大剂量甲氨蝶呤冲击疗法预防性治疗髓外白血病。在临床中，大剂量甲氨蝶呤常与其他药物合用，如抗生素、质子泵抑制剂、非甾体抗炎药等可能导致甲氨蝶呤体内药代动力学改变，影响甲氨蝶呤用药安全性。近年来多个案例报道甲氨蝶呤排泄延迟与联合用药有一定的相关性，本文对大剂量甲氨蝶呤联合用药情况及影响机制进行综述分析，提高甲氨蝶呤临床应用的安全性。 |

| Methotrexate (methotrexate,MTX) is a dihydrofolate reductase inhibitor, clinical high-dose of methotrexate shock therapy for the preventive treatment of extractrexary leukemia. In clinical applications, Large doses of methotrexate are often used with other drugs, such as antibiotics, proton pump inhibitors, and non-steroidal anti-inflammatory drugs that may lead to pharmacokinetic changes in methotrexate and affect the safety of methotrexate medication. In recent years, several cases have reported a correlation between delayed methotrexate excretion and co-administration. In this paper, we review and analyse the situation of high-dose methotrexate co-administration and the mechanism of influence to improve the safety of methotrexate clinical application. |
