载甲状旁腺激素相关蛋白的聚乳酸羟基乙酸微球的制备、形态表征及生物相容性的研究

摘要:
目的:制备聚乳酸羟基乙酸-甲状旁腺激素相关蛋白复合微球(PLGA-PTHrP)并观察其表征。方法:采用复乳溶剂挥发法制备空白聚乳酸羟基乙酸复合物微球(PLGA),通过表面浸提法将不同浓度的甲状旁腺激素相关蛋白(PTHrp)装载到PLGA微球。冷冻干燥法制备出负载了有效浓度PTHrP的PLGA微球。扫描电镜观察PLGA微球形态并且计算微球粒径; 利用BCA试剂盒、酶标仪检测PLGA-PTHrP的蛋白释放量,绘制释放曲线。将PLGA-PTHrP与大鼠全骨髓细胞共培养,细胞计数试剂盒法(CCK-8)检测复合微球对细胞增殖分化的影响。结果:采用水/油/水(W/O/W)复乳溶剂挥发法制备得到的空载PLGA微球呈球形或类球形,表面较平整,平均粒径56.7±7.2微米(μm)。载蛋白微球的体外蛋白质释放实验显示:蛋白质的释放速率在前100小时内较快,释放时间可持续达300小时。最终,载蛋白微球释放的蛋白质占蛋白质总量的87.3%。CCK-8结果显示:PLGA-PTHrP对大鼠全骨髓细胞增殖活力无明显影响。 结论:通过复乳溶剂挥发法能制备出表面较平整,粒径较均匀的PLGA空白微球,使用表面浸提法以及冷冻干燥法能制备出成功有效的PLGA-PTHrP缓释体系,该缓释体系能在一定时间范围内实现PTHrP的缓释作用。

Preparation, characterization and biocompatibility of polylactide-co-glycolide microspheres loaded with parathyroid hormone-related protein(PLGA-PTHrP)

Abstract:
Objective: Prepare polylactide-co-glycolide (PLGA) microspheres and explore the characterization of PLGA microspheres loaded with parathyroid hormone-related protein (PTHrP) Methods: The empty PLGA microspheres were prepared by double emulsion-solvent evaporation method. PTHrP were loaded on the empty PLGA microspheres by soaking empty PLGA microspheres in the different concentrations of PTHrP solution. PLGA microspheres loaded with effective concentration of PTHrP were prepared by freeze-drying method. The surface morphology and average particle size of PLGA microspheres were observed by SEM scanning electron microscopy. BCA kit and microplate reader were used to detect the protein release curve. PLGA-PTHrP were co-cultured with rat whole bone marrow cells, then Cell Counting Kit-8(CCK-8) were used to detect the effect of PLGA-PTHrP MPs on the osteogenic ability of whole bone marrow cells. Results: The empty PLGA microspheres made by W/O/W double emulsion-solvent evaporation method were spherical or kind of spherical with round surface, and its average size were 56.7±7.2 μm. The in vitro PTHrP release experiment showed that The release rate of protein were faster in the first 100 hours. The total release time lasted for 300 hours.PLGA-PTHrP MPs could release about 87.3% protein of the total. CCK-8 showed PLGA-PTHrP MPs had no significantly effect on the proliferation activity of whole bone marrow cells. CONCLUSION: The PLGA empty microspheres can be successfully made by W/O/W double emulsion-solvent evaporation method, and PLGA-PTHrP sustained-release system made by freeze-drying method maintain a sustained release effect.
