下调miR-27a减轻心肌缺血再灌注大鼠急性肺损伤

摘要:
目的 评价下调miR-27a对大鼠心肌缺血再灌注后所引起急性肺损伤的保护作用及其机制。方法 将40只8-12周成年大鼠随机分为4组(n=10):假手术组(SP组)、模型组(I/R组)、miR-27a抑制剂组(anta-27a+I/R组)、空白对照组(NC+I/R组)。结扎冠脉左前降支构建大鼠心肌缺血再灌注所致急性肺损伤模型。anta-27a+I/R组与NC+I/R组大鼠于造模前连续3天尾静脉分别注射antagomir-27a、antagomir-NC。I/R组大鼠结扎冠脉左前降支30 min,再灌注120 min。SP组大鼠只穿线不接扎。再灌注120 min后取血,迅速取肺组织,右肺测定肺湿干重比(W/D),左肺上叶HE染色,采用光镜观察肺组织病理改变,比色法检测肺组织中丙二醛(MDA)和超氧化物歧化酶(SOD)水平。ELISA法检测肺组织及血清中白介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)。结果 与SP组比较,I/R组、anta-27a+I/R组、NC+I/R组大鼠肺组织病理学形态损伤严重,肺W/D比值升高,肺组织MDA水平明显增加,SOD活性明显降低,肺组织及血清中IL-6和TNF-α的含量明显增加(P<0.05);与I/R组比较,anta-27a+I/R组大鼠肺组织病理损伤程度有所减轻,肺W/D有所下降,肺组织MDA水平下降,SOD活性增加,肺组织及血清中IL-6和TNF-α的含量下降(P<0.05)。结论 下调miR-27a表达可以通过减轻炎症反应与氧化应激缓解大鼠心肌缺血再灌注后所引起的急性肺损伤。

Down-regulation of miR-27a alleviates acute lung injury in rats by myocardial ischemia-reperfusion

Abstract:
Objective To evaluate the protective effect of miR-27a down-regulation on acute lung injury in rats by myocardial ischemia-reperfusion. Methods Forty adult rats of 8-12 weeks were divided into four groups:sham surgery group (SP group), model group (I/R group), miR-27a inhibitor group (anta-27a+ I/R group), and blank control group (NC+I/R group). The rats modle of acute lung injury induced by MI/R was esdablished by ligation of coronary artery branch. Rats in the anta-27a+I/R and NC+I/R groups were intravenous injected with antagomir-27a and antagomir-NC for 3 consecutive days before molding. Rats in IR group subject to reperfusion for 120 min after 30 min ischemia. Rats in the SP group underwent thoracotomy without artery ligation. After 120 min of reperfusion, lung tissue was collected quickly, the ratio of wet and dry weight (W/D) was measured, and lung organization pathological changes were observed under the HE staining. SOD and MDA levels in lung tissue were determined by colorimetry. Interleukin-6(IL-6) and tumor necrosis factor-α(TNF-α) in serum and lung tissue were measured by ELISA. Results Compared with the SP group, rats in the I/R , the NC+I/R and anta-27a+I/R groups had severe lung injury, including increased lung W/D ratio, increased MDA level, decreased SOD activity, and increased serum and lung tissue IL-6 and TNF-α(P<0.05). Compared with the I/R group, the pathological injury of lung tissue was reduced, in anta-27a+I/R group, lung W/D ratio was decreased, MDA level was decreased, SOD activity was raised and serum and lung tissue IL-6 and TNF-α were decreased(P<0.05). Conclusion Low expression of miR-27a can alleviate the acute lung injury caused by myocardial ischemia and reperfusion in rat by reducing inflammation response and oxidative stress.
