芒果叶提取物体外抗HIV-1活性研究

摘要:
目的 研究传统中草药芒果叶提取物的体外细胞毒性和抗HIV-1活性,为治疗艾滋病的传统中医药资源的研究与开发提供参考依据。方法 通过ATP法评估药物对TZM-bl细胞和MT-2细胞的细胞毒性作用;构建TZM-bl-HIV-1IIIB、MT2-HIV-1IIIB两种HIV-1细胞感染模型,通过荧光素酶活性检测试剂检测病毒活性从而评估药物抗HIV-1活性,同时观察MT2-HIV-1IIIB细胞系统中药物对细胞病变效应(CPE)的抑制情况。通过计算药物对细胞的半数毒性浓度(CC50)从而确定药物的体外细胞毒性,计算药物的半数抑制浓度(IC50)从而确定药物对HIV-1复制的抑制活性,并根据CC50和IC50计算药物的选择指数(SI)。结果 在不同细胞感染模型中,芒果叶提取物均具有抗HIV-1活性,且呈现剂量依赖性。在TZM-bl-HIV-1IIIB模型中,CC50为320.00±29.44 μg·mL-1,IC50为8.86±0.26μg·mL-1,SI为36.14。在MT-2-HIV-1IIIB模型中,CC50为174.13±22.36μg·mL-1, IC50为15.23±9.99μg·mL-1,SI为11.44,且抑制HIV-1诱导的细胞病变效应作用较为显著。结论 传统中草药芒果叶提取物的体外细胞毒性小,抗HIV-1活性显著,具有潜在的开发利用价值。

Abstract:
Objective In this study, we aimed to reseach the cytotoxicity and anti-HIV-1 activity of mango(Mangifera indica L.) leaves extract in vitro and provide reference basis for the development of traditional Chinese medicine resources for AIDS treatment. Methods The cytotoxicity of the extract on TZM-bl and MT2 cells was evaluated by ATP assay. TZM-bl-HIV-1IIIB and MT2-HIV-1IIIB cell infection models were established to evaluate the anti-HIV-1 activity by detecting the expression of luciferase activity. Meanwhile, the inhibition of cytopathic effect (CPE) in MT2-HIV-1IIIB cell infection model was observed. Furthermore, the half cytotoxic concentration (CC50) was calculated to determine the cytotoxicity of the extract in vitro. The half inhibitory concentration (IC50) was calculated to determine the inhibitory activity of the extract on HIV-1 replication. And the drug selection index (SI) was calculated according to CC50 and IC50. Results Our results revealed that Mango(Mangifera indica L.) leaves extract showed dose-dependent anti-HIV-1 activity in different cell infection models. In TZM-bl-HIV-1IIIB model, the CC50 was 320.00±29.44 μg·mL-1, the IC50 was 8.86±0.26 μg·mL-1, and SI was 36.14. In MT-2-HIV-1IIIB model, the CC50 was 174.13±22.36μg·mL-1, the IC50 was 15.23±9.99μg·mL-1, and the SI was 11.44. Moreover, the cytopathic effect induced by HIV-1 was inhibited significantly by the extract in MT-2-HIV-1ⅢB model. Conclusion The traditional Chinese herbal medicine mango(Mangifera indica L.) leaves extract shows low cytotoxicity in vitro and remarkable anti-HIV-1 activity, which has potential development and utilization value.
