[摘要] 目的 探讨一种高效、经济、稳定的肝星状细胞的分离方法,为肝纤维化研究提供细胞模型。方法 应用链霉蛋白酶、胶原酶灌流,以Nycodenz为分离介质,采用密度梯度离心法分离大鼠的肝星状细胞。结果 肝星状细胞的获得率为(3.2±0.67)×107/只、存活率为90%以上。结论 该法是一种较理想的分离肝星状细胞的方法。 |

[Abstract] Objective To develop a productive,inexpensive and stable method for the isolation and culture of rat hepatic stellate cells (HSCs) as a cell model for the studies of hepatic fibrosis.Methods Rat HSCs were isolated by liver perfusion with pronase and collagenase followed by density gradient centrifugation with Nycodenz. Results The harvest rate of hepatic stellate cells was (3.2±0.67)×107 cells per liver and viability rate was more than 90%.Conclusion This method is an ideal option for the isolation and culture of rat HSCs. |
