[摘要] 目的 探讨柠檬酸钠(sodium citrate,CT)与三氧化二砷(As2O3,AS)联合应用对胃癌SGC-7901细胞凋亡及作用机制。方法 用sodium citrate(终浓度为5 mmol/L,CT5)和As2O3(终浓度依次为5 μmol/L,AS5)处理体外传代培养的胃癌SGC-7901细胞株。应用DAPI细胞核染色和流式细胞仪检测细胞凋亡和细胞周期的变化;RT-PCR法观察抗凋亡相关基因bcl-2表达的变化。结果 sodium citrate与As2O3联合应用相对于单用sodium citrate或As2O3可显著提高诱导胃癌细胞凋亡率(P<0.05);与空白组相比,用药后G0/G1期细胞比例下降,G2/M期细胞比例上升,细胞周期阻滞于G2/M期,抗凋亡基因bcl-2表达明显下降。结论 sodium citrate与As2O3联合应用具有协同抗胃癌细胞的作用,其机制可能与细胞周期阻滞、增强诱导胃癌细胞凋亡以及调控凋亡相关基因的表达有关。 |

[Abstract] Objective To study the anti-tumor effects of sodium citrate combined with arsenic trioxide on human gastric cancer cell line SGC-7901 in vitro and to explore its possible mechanism.Methods The passaged and cultured gastric cancer cell line SGC-7901 in vitro were treated with sodium citrate ( final concentration was 5mmol/L,CT5) and(or)As2O3 (final concentration were 5 μmol/L,AS5).Cell proliferation was observed by fluorescent inversive microscopy; Cell apoptosis was explored by DAPI fluorescent stain, then the cell apoptosis rate and phases of cell cycle were determined by flow cytometry. The expression of apoptosis-related genes of bcl-2 was observed by RT-PCR.Results Sodium citrate combined with As2O3 had a stronger inhibition effect on human gastric cancer cell line SGC-7901 than sodium citrate or As2O3(P<0.05). Compared with blank group,the cell cycle distribution was altered and the ration of G0/G1 phase cell decreased and G2/M phase increased (P<0.05). After intervened by sodium citrate combined with As2O3,the expression of bcl-2 decreased(P<0.05).Conclusion Sodium citrate combined with As2O3 has the synergistic effects on gastric cancer cell line SGC-7901,and the mechanisms may be related to cell cycle blocking and apoptosis-inducing effects through regulating the expression of the related genes. |
