体外冲击波联合仙桃草治疗骨不连对骨痂中BMP-2和VEGF mRNA表达的影响

摘要:
[摘要] 目的 探讨体外冲击波联合中草药仙桃草疗法对兔桡骨骨不连骨痂中骨形态发生蛋白-2(BMP-2)、血管内皮生长因子(VEGF)表达的影响。方法 制作120只新西兰大白兔兔桡骨骨不连模型,按随机数字表方法分为A、B、C、D四组,A组为冲击波联合仙桃草治疗组,B组为仙桃草治疗组,C组为冲击波治疗组,D组为对照组。体外冲击波治疗方法:在骨不连两断端部位冲击,冲击量为800次/min,焦点能量0.5 mj/mm2,频率为70~80次/min。于治疗后不同时相点(48 h、1、2、4、6、12周)取材,比较各组标本BMP-2和VEGF的免疫组化表达,并作统计分析。结果 (1)各时相点骨痂中BMP-2的表达:48 h各组相比差异无统计学意义。1、2、4、6、12周各组不同时相点比较,A、C组与D组比较差异有统计学意义(P<0.05),B组与D组比较差异无统计学意义(P>0.05),且A组与B、C组比较差异同样有统计学意义(P<0.05)。(2)各时相点骨痂中VEGF的表达:A、B、C组与D组比较差异均有统计学意义(P<0.05),且A组与B、C组比较差异也有统计学意义(P<0.05)。结论 体外冲击波联合仙桃草疗法可以同时上调BMP-2、VEGF表达,从而刺激成骨,共同促进骨不连愈合。

Effects of extracorporeal shock wave combined with veronica peregrina on the expressions of BMP-2 and VEGF mRNA in the treatment of fracture nonunion

Abstract:
[Abstract] Objective To investigate the effects of extracorporeal shock wave combined with veronica peregrina on BMP-2 and VEGF mRNA expressions in the callus of fracture nonunion in rabbits, and to explore its mechanism.Methods The model of radius fracture nonunion was made in 120 New Zealand white rabbits, which were then randomly divided into four groups: group A in the treatment of extracorporeal shock wave combine with veronica peregrina, group B in the treatment of veronica peregrina, group C in the treatment of extracorporeal shock wave and group D as the model control. The tested animals were treated with extracorporeal shock wave, and the two ends of the nonunion frocture were shocked 800 times at 0.5 mj/mm2 and a frequency of 70~80 times/minute. The samples were collected in each group at the end of 48 hours, 1, 2,4, 6, 12 weeks respectively after the treatment. Immunohisto chemistry stain was performed to examinate the expressions of BMP-2 and VEGF mRNA in the callus.Results (1)There were no significant differences in the expressions of BMP-2 mRNA in callus at each time point among the four groups 48 hours after the treatment. At the end of 1, 2, 4, 6 and 12 weeks, group A and group C had significant difference compared with group D(P<0.05). Group B had no significant difference compared with group D(P>0.05). Group A had significant difference compared with group B and C(P<0.05).(2)Group A,group B and group C had significant differences in the expression of VEGF mRNA in callus at each time point compared with group D(P<0.05). Group A had significant difference compared with group B and group C(P<0.05).Conclusion The treatment of extracorporeal shock wave combined with veronica peregrina can upregulate both the expressions of BMP-2 and VEGF mRNA, stimulate osteogenesis and improve the healing of fracture nonunion.
