钙黏蛋白13表达下调与膀胱癌生长的关联性研究

摘要:
[摘要] 目的 探讨钙黏蛋白13(cadherin-13,CDH13)表达下调与膀胱癌生长关联性及其对磷脂酰肌醇3激酶(phosphatidylinositol 3 kinase,PI3K)/磷酸化蛋白激酶B(phosphorylated protein kinase B,p-Akt)信号通路的影响。方法 应用膀胱癌5637细胞建立裸鼠成瘤模型,比较不同时间点正常对照组(以1×PBS处理5637细胞,n=7)、阴性对照组(以shRNA质粒转染5637细胞,n=7)和实验组(以CDH13 shRNA质粒转染5637细胞,n=7)的肿瘤体积。采用Western blot法检测肿瘤组织中CDH13、PI3K及p-Akt蛋白的表达情况,并比较这三种蛋白在三组间的表达差异。结果 在实验期间,三组裸鼠的肿瘤体积均呈增长趋势,实验组肿瘤体积增大最快,三组变化幅度差异有统计学意义(P<0.05)。在第10天、第13天和第16天,实验组裸鼠肿瘤体积显著大于阴性对照组和正常对照组(P<0.05),而阴性对照组肿瘤体积显著大于正常对照组(P<0.05)。Western blot实验结果显示,实验组CDH13蛋白表达水平显著低于阴性对照组和正常对照组(P<0.05),而PI3K与p-Akt蛋白的表达水平则显著高于阴性对照组和正常对照组(P<0.05)。结论 CDH13表达下调可能通过PI3K/p-Akt信号通路促进膀胱癌的发生。

Study on the correlation between down-regulation of cadherin-13 expression and bladder cancer growth

Abstract:
[Abstract] Objective To investigate the correlation between down-regulation of cadherin-13(CDH13) expression and the growth of bladder cancer, and its effects on phosphatidylinositol 3 kinase(PI3K)/phosphorylated protein kinase B(p-Akt)signal pathways. Methods Nude mouse tumor formation models were established using bladder cancer 5637 cells, and tumor volume was compared among the normal control group(5637 cells treated with 1×PBS, n=7), the negative control group(transfected 5637 cells with shRNA plasmid, n=7) and the experimental group(transfected 5637 cells with CDH13 shRNA plasmid, n=7) at different time points. Western blot was used to detect the expressions of CDH13, PI3K and p-Akt proteins in the tumor tissues, and the expressions of the three proteins were compared among the three groups. Results During the experimental period, the tumor volume showed an increasing trend in the nude mice of the three groups, and the tumor volume of the experimental group increased fastest, and the change magnitude was statistically significant among the three groups(P<0.05). On the 10th day, the 13th day, and the 16th day, the tumor volume of the nude mice in the experimental group was significantly larger than that in the negative control group and the normal control group(P<0.05), while the tumor volume of the negative control group was significantly larger than that of the normal control group(P<0.05). Western blot results showed that the level of CDH13 expression in the experimental group was significantly lower than that in the negative control group and the normal control group(P<0.05), while the expression levels of PI3K and p-Akt proteins in the experimental group were significantly higher than those in the negative control group and the normal control group(P<0.05). Conclusion Down-regulation of CDH13 expression may promote the occurrence of bladder cancer through the PI3K / p-Akt signaling pathways.
