乳腺癌中PBK/TOPK mRNA表达的临床意义及功能预测

[摘要] 目的 通过生物信息学分析PDZ结合激酶/T-淋巴因子激活的杀伤细胞来源的蛋白激酶(PBK/TOPK)在乳腺癌(BRCA)中的表达及临床意义,并预测其潜在的分子通路和生物学功能。方法 挖掘Oncomine数据库分析PBK/TOPK信使核糖核酸(mRNA)在不同癌症中的表达情况。下载癌症基因组图谱(TCGA)数据库BRCA和正常乳腺组织的RNA测序数据和临床信息,分析PBK/TOPK mRNA表达与临床特征的相关性。应用Kaplan-Meier Plotter数据库进行生存分析。通过筛选PBK/TOPK相关基因,并进行功能富集分析,预测PBK/TOPK在BRCA中的潜在分子通路和生物学功能。结果 PBK/TOPK基因在BRCA组织中的表达显著高于正常乳腺组织(P<0.05),并与雌激素受体(ER)、孕激素受体(PR)、三阴性乳腺癌(TNBC)和绝经状态存在关联性(P<0.05)。生存分析结果显示,PBK/TOPK基因高表达提示BRCA预后不良(P<0.05)。功能富集分析显示PBK/TOPK相似基因主要涉及BRCA细胞分裂、细胞周期调节和微管细胞骨架形成等生物功能,参与BRCA细胞周期等信号通路。结论 PBK/TOPK mRNA在BRCA组织中高表达,与BRCA患者预后不良密切相关,可能是BRCA潜在的生物标志物和治疗靶标。

Clinical significance and function prediction of mRNA expression of PBK/TOPK in breast cancer

Abstract:
[Abstract] Objective To analyze the expression of PDZ-binding kinase/T-LAK cell-originated protein kinase(PBK/TOPK) in breast cancer(BRCA) by bioinformatics and its clinical significance, and to predict its potential molecular pathways and biological functions. Methods Oncomine database was mined to analyze the expressions of PBK/TOPK messenger ribonucleic acid(mRNA) in different cancers. Ribonucleic acid(RNA) sequencing data and clinical information of BRCA tissue and normal breast tissue were downloaded from the Cancer Genome Atlas(TCGA) database to analyze the correlation between PBK/TOPK mRNA expression and clinical characteristics, and survival analysis was performed using the Kaplan-Meier Plotter database. The related genes of PBK/TOPK were screened, and functional enrichment analysis was performed to predict the potential molecular pathways and biological functions of PBK/TOPK in BRCA. Results The expression of PBK/TOPK gene in BRCA tissue was significantly higher than that in normal breast tissue(P<0.05), and was correlated with estrogen receptor(ER), progesterone receptor(PR), triple negative breast cancer(TNBC) and menopause status(P<0.05). Survival analysis showed that the high expression of PBK/TOPK gene indicated a poor prognosis of BRCA(P<0.05). Functional enrichment analysis showed that similar genes of PBK/TOPK were mainly involved in biological functions such as cell division, cell cycle regulation and microtubule cytoskeleton formation in BRCA, and involved in BRCA cell cycle and other signaling pathways. Conclusion PBK/TOPK mRNA is highly expressed in BRCA tissues, which is related to poor prognoses of BRCA patients. PBK/TOPK mRNA can be used as a potential biomarker and therapeutic target for BRCA.
