lncRNA TUG1和miR-26a在子痫前期胎盘组织中的表达水平及其临床意义

摘要:
[摘要] 目的 探讨子痫前期(PE)患者胎盘组织中长链非编码RNA牛磺酸上调基因1(lncRNA TUG1)和微小RNA-26a(miR-26a)的表达水平,并分析两者在PE发生、发展中的作用。方法 选择2019年5月至2021年5月株洲市中心医院产科收治的新发PE患者100例(PE组),其中轻度PE 49例(轻度PE组),重度PE 51例(重度PE组)。以年龄、胎龄为匹配条件,纳入同期健康孕妇100名作为对照组。通过实时荧光定量PCR(qRT-PCR)检测研究对象胎盘组织中lncRNA TUG1和miR-26a的表达水平。通过Pearson相关分析探讨lncRNA TUG1、miR-26a表达水平与PE患者临床指标的相关性。结果 轻度PE组、重度PE组胎盘组织的lncRNA TUG1表达水平低于对照组,miR-26a表达水平高于对照组,差异有统计学意义(P<0.05)。与轻度PE组比较,重度PE组胎盘组织lncRNA TUG1表达水平更低,miR-26a表达水平更高,差异有统计学意义(P<0.05)。Pearson相关性分析结果显示,PE患者胎盘组织中lncRNA TUG1表达水平与收缩压、舒张压和尿蛋白水平呈负相关(P<0.05);miR-26a表达水平与收缩压、舒张压和尿蛋白水平呈正相关(P<0.05);lncRNA TUG1表达水平与miR-26a表达水平呈负相关(P<0.05)。结论 lncRNA TUG1在PE胎盘组织中呈低表达,而miR-26a呈高表达,其与PE严重程度具有关联性。lncRNA TUG1可能通过靶向结合miR-26a参与PE的发生、发展。

Abstract:
[Abstract] Objective To investigate the expression levels of long non-coding RNA taurine up-regulated gene 1(lncRNA TUG1) and microRNA-26a(miR-26a) in placental tissues of the patients with preeclampsia(PE), and to analyze their roles in the occurrence and development of PE. Methods One hundred newly diagnosed PE patients(the PE group) admitted to the Department of Obstetrics of Zhuzhou Municipal Central Hospital from May 2019 to May 2021 were selected, including 49 patients with mild PE(the mild PE group) and 51 patients with severe PE(the severe PE group). One hundred healthy pregnant women were included in the control group during the same period based on the matching age and gestational age. The expression levels of lncRNA TUG1 and miR-26a in the placental tissues of the study subjects were detected by quantitative real-time polymerase chain reaction(qRT-PCR). Pearson correlation analysis was used to investigate the correlation between the expression levels of lncRNA TUG1 and miR-26a and the clinical indicators of the PE patients. Results The expression levels of lncRNA TUG1 in the placental tissues in the mild PE group and the severe PE group were lower than those in the control group, and the expression levels of miR-26a in the mild PE group and the severe PE group were higher than those in the control group, and the differences were statistically significant(P<0.05). Compared with those in the placental tissues of the mild PE group, the expression level of lncRNA TUG1 was lower and the expression level of miR-26a was higher in the placental tissues of the severe PE group, and the differences were statistically significant(P<0.05). The results of Pearson correlation analysis showed that the expression level of lncRNA TUG1 in the placental tissues of the PE patients was negatively correlated with systolic blood pressure, diastolic blood pressure and urinary protein level(P<0.05), and that the expression level of miR-26a was positively correlated with systolic blood pressure, diastolic blood pressure and urinary protein level(P<0.05). The expression level of lncRNA TUG1 was negatively correlated with that of miR-26a(P<0.05). Conclusion lncRNA TUG1 is low expressed in PE placental tissues, while miR-26a is highly expressed, which is associated with the severity of PE. lncRNA TUG1 may participate in the occurrence and development of PE by targeting miR-26a.
