系统性红斑狼疮患者外周血AC007278.2、MX2表达水平与疾病活动度及补体水平的关联性分析

摘要:
[摘要] 目的 分析系统性红斑狼疮(SLE)患者外周血长链非编码RNA AC007278.2、MX动力蛋白样GTP酶2(MX2)表达水平与疾病活动度、补体水平的关联性。方法 招募2018年11月至2020年2月武汉亚心总医院及中部战区总医院收治的SLE患者168例(SLE组)。根据患者入院24 h内进行SLE疾病活动指数-2000(SLEDAI-2000)评分,其中疾病活动度为无活动者42例,轻度活动者45例,中度活动者42例,重度活动者39例。招募同期健康人群80名作为对照组。比较不同组的临床资料,采用Pearson相关分析探讨AC007278.2、MX2水平与补体C3、C4水平及疾病活动度的相关性,采用多元线性回归分析影响SLE疾病活动度的因素。结果 SLE组AC007278.2、MX2水平高于对照组,补体C3、C4及白细胞(WBC)、淋巴细胞、血小板(PLT)、血红蛋白(Hb)水平低于对照组,差异有统计学意义(P<0.05)。Pearson相关分析结果显示,SLE患者AC007278.2、MX2水平与补体C3、C4水平呈负相关(P<0.05),与SLEDAI-2000评分呈正相关(P<0.05)。多元线性回归分析结果显示,AC007278.2(β=0.410)、MX2(β=0.512)与SLE疾病活动度呈正关联(P<0.05),补体C3(β=-0.362)、C4(β=-0.528)与SLE疾病活动度呈负关联(P<0.05)。结论 SLE患者外周血AC007278.2、MX2表达水平升高,与SLE疾病活动度及补体C3、C4具有关联性。

Analysis on the correlation of expression levels of AC007278.2 and MX2 in the peripheral blood of the systemic lupus erythematosus patients with disease activity and complement level

Abstract:
[Abstract] Objective To analyze the correlation of expression levels of long non-coding RNA AC007278.2 and MX dynamin-like GTPase 2(MX2) in the peripheral blood of the systemic lupus erythematosus(SLE) patients with disease activity and complement level. Methods One hundred and sixty-eight SLE patients(SLE group) admitted to Wuhan Asia General Hospital and Central Command General Hospital of the Chinese People′s Liberation Army from November 2018 to February 2020 were recruited. According to SLE disease activity index-2000(SLEDAI-2000) scores of the patients within 24 hours after admission, 42 patients were inactive, and 45 patients were mildly active, and 42 patients were moderately active, and 39 patients were severely active in disease activity. Eighty healthy people at the same period were recruited as control group. The clinical data of different groups were compared, Pearson correlation analysis was used to explore the correlation between AC007278.2 and MX2 levels and complement C3 and C4 levels as well as disease activity, and multiple linear regression was used to analyze the factors affecting SLE disease activity. Results The levels of AC007278.2 and MX2 in the SLE group were higher than those in the control group, and the levels of complement C3 and C4, white blood cells(WBC), lymphocytes, platelets(PLT), and hemoglobin(Hb) in the SLE group were lower than those in the control group, and the differences were statistically significant(P<0.05). The results of Pearson correlation analysis showed that AC007278.2 and MX2 levels were negatively correlated with complement C3 and C4 levels in the SLE patients(P<0.05), and were positively correlated with SLEDAI-2000 scores(P<0.05). The results of multiple linear regression analysis showed that AC007278.2(β=0.410) and MX2(β=0.512) were positively correlated with SLE disease activity(P<0.05), while complement C3(β=-0.362) and complement C4(β=-0.528) were negatively correlated with SLE disease activity(P<0.05). Conclusion The expression levels of AC007278.2 and MX2 in the peripheral blood of SLE patients are elevated and are associated with SLE disease activity and complement C3 and C4.
