直肠癌IER3基因表达与临床病理特征及预后的研究

【】目的 检测直肠癌组织中IER3基因及蛋白的表达情况,分析其与直肠癌患者临床病理特征及预后的关系。方法 (1)应用免疫组织化学技术检测IER3 蛋白表达、qPCR检测IER3 mRNA表达、CanPatrol? 技术检测术前循环血肿瘤细胞(CTCs),通过数据分析评估IER3基因的表达与直肠癌病理特征、术前CTCs数量及亚型的相关性,并对预后进行随访及生存分析。结果 IER3基因mRNA和蛋白表达在直肠癌组织中显著高于对应的癌旁正常组织中的表达水平(P<0.05 )。IER3的mRNA表达差异与肿瘤大小正相关(P<0.05),与脉管侵犯有相关性趋势,但无统计学差异(P=0.06),而与性别、年龄、T分期、N分期、TNM分期、神经侵犯无相关性。IER3蛋白表达与术前混合型CTCs、术前E型CTCs数量有相关性(P<0.05)。术前CTCs数量与血液CEA水平有相关性(P<0.05)。通过生存分析,直肠癌组织IER3蛋白和 mRNA表达差异与直肠癌患者预后无直接相关性(P>0.05)。结论 本研究显示,直肠癌组织中IER3基因在mRNA和蛋白表达水平显著上调,并与肿瘤大小和特定CTCs亚型等临床病理特征存在相关性,提示其在CTCs适应性生存中可能发挥作用,但具体机制仍需进一步研究。

Expression of the IER3 Gene in Rectal Cancer and Its Association with Clinicopathological Characteristics and Prognosis

Abstract:
Objective: To detect the expression of the IER3 gene and its protein in rectal cancer tissues, and to analyze its relationship with clinicopathological characteristics and prognosis in patients with rectal cancer.Methods: (1) Immunohistochemistry was used to detect IER3 protein expression, qPCR was used to detect IER3 mRNA expression, and the CanPatrol? technique was employed to detect preoperative circulating tumor cells (CTCs). Data analysis was performed to evaluate the correlation between IER3 expression and rectal cancer pathological features, preoperative CTC count, and CTC subtypes. Prognosis was assessed through follow-up and survival analysis.Results: The mRNA and protein expression levels of the IER3 gene were significantly higher in rectal cancer tissues compared to adjacent normal tissues (P < 0.05). The differential expression of IER3 mRNA was positively correlated with tumor size (P < 0.05) and showed a trend toward correlation with vascular invasion, though this was not statistically significant (P = 0.06). No significant correlations were observed with gender, age, T stage, N stage, TNM stage, or nerve invasion. IER3 protein expression was correlated with the counts of preoperative mixed-type CTCs and preoperative epithelial-type (E-type) CTCs (P < 0.05). Preoperative CTC count was correlated with CEA levels? (P < 0.05). Survival analysis indicated no direct correlation between IER3 protein or mRNA expression in rectal cancer tissues and patient prognosis (P > 0.05).Conclusion: This study demonstrates that IER3 gene expression is significantly upregulated at both the mRNA and protein levels in rectal cancer tissues and correlates with clinicopathological features such as tumor size and specific CTC subtypes. These findings suggest a potential role for IER3 in the adaptive survival of CTCs, although the underlying mechanisms require further investigation.
