| 分子诊断日益成为病原体检测的金标准。核酸扩增结合成簇规律间隔的短回文重复序列(Clustered Regularly Interspaced Short Palindromic Repeats, CRISPR)的分子检测系统具有高特异性识别、可编程、灵敏度高和操作便携等特性而被认为新一代即时分子诊断技术。CRISPR一管法是将核酸扩增和CRISPR检测集成于单一反应体系,简化了操作流程,大大降低了气溶胶污染的可能性,具备理想即时检测(POCT)设备的特性,因此备受研究者关注。近年来,基于多种生物传感器进行最终信号测量的核酸扩增——CRISPR的一管法系统已经被开发出来,将整个测量程序的性能及便捷程度朝着实用化和现场化应用不断推进,使其在病原体快速检测、食品安全和环境监测领域颇具应用潜力。本文就多种基于核酸扩增-CRISPR一管法的生物传感器及应用场景进行综述,为即时分子检测方法研究及未来病原体防治提供新的思路和策略。 |

| Molecular diagnostics have increasingly become the gold standard for pathogen detection. Detection systems that integrate nucleic acid amplification with Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR) technology are regarded as a new generation of point-of-care molecular diagnostics due to their high specificity, programmability, sensitivity, and operational portability. CRISPR-based one-pot assays, which integrate nucleic acid amplification and CRISPR-mediated detection into a single reaction system, significantly simplify operational procedures and substantially reduce the risk of aerosol contamination, thereby exhibiting the key attributes of ideal point-of-care testing (POCT) platforms and attracting considerable research interest. In recent years, one-pot nucleic acid amplification–CRISPR systems incorporating diverse biosensors for terminal signal readout have been developed, continuously advancing overall assay performance and usability toward practical and field-deployable applications. These systems demonstrate substantial potential in rapid pathogen detection, food safety monitoring, and environmental surveillance. This review summarizes various biosensors integrated with nucleic acid amplification–CRISPR one-pot systems and their application scenarios, aiming to provide new insights and strategic perspectives for the development of point-of-care molecular diagnostics and future pathogen prevention and control. |
