芒果叶提取物体外抗HIV-1活性研究

[摘要] 目的 探讨芒果叶提取物的体外抗人类免疫缺陷病毒1(HIV-1)活性,为研究与开发传统中医药资源治疗获得性免疫缺陷综合征(AIDS)提供参考。方法 通过三磷酸腺苷(ATP)法评估干预药物对TZM-bl细胞和MT-2细胞活性的影响。构建TZM-bl-HIV-1ⅢB、MT-2-HIV-1ⅢB两种细胞-病毒感染模型,通过荧光素酶活性检测试剂检测病毒活性,评估干预药物的抗HIV-1活性。观察MT-2-HIV-1ⅢB细胞系统中芒果叶提取物对细胞病变效应(CPE)的抑制情况。计算芒果叶提取物的半数毒性浓度(CC50)、半数抑制浓度(IC50)和选择指数(SI)。结果 在TZM-bl-HIV-1ⅢB和MT-2-HIV-1ⅢB两种细胞-病毒感染模型中,芒果叶提取物均显示出抗HIV-1活性,且呈现剂量依赖性。在TZM-bl-HIV-1ⅢB模型中,芒果叶提取物的CC50为(320.00±29.44)μg/ml,IC50为(8.86±0.26)μg/ml,SI为36.14。在MT-2-HIV-1ⅢB模型中,芒果叶提取物CC50为(174.13±22.36)μg/ml,IC50为(15.23±9.99)μg/ml,SI为11.44。结论 芒果叶提取物的体外细胞毒性小,抗HIV-1活性显著,具有潜在的开发利用价值。

[Abstract] Objective To investigate the in-vitro activity of Mango(Mangifera indica L.) leaf extract against human immunodeficiency virus 1(HIV-1), and to provide reference for the research and development of Traditional Chinese Medicine resources for the treatment of acquired immune deficiency syndrome(AIDS). Methods The effects of intervention drugs on the activity of TZM-bl cells and MT-2 cells were evaluated by adenosine triphosphate(ATP) assay. Two cell-virus infection models of TZM-bl-HIV-1ⅢB and MT-2-HIV-1ⅢB were constructed, and the virus activity was detected by luciferase activity detection reagent, so as to evaluate the anti-HIV-1 activity of intervention drugs. The inhibition of Mango leaf extract on cytopathic effect(CPE) in MT-2-HIV-1ⅢB cell system was observed. Half toxic concentration(CC50), half inhibitory concentration(IC50) and selection index(SI) of Mango leaf extract were calculated. Results In the TZM-bl-HIV-1ⅢB and MT-2-HIV-1ⅢB cell-virus infection models, Mango leaf extract showed anti-HIV-1 activity in a dose-dependent manner. In the TZM-bl-HIV-1ⅢB model, the CC50, IC50 and SI of Mango leaf extract were (320.00±29.44)μg/ml, (8.86±0.26)μg/ml and 36.14, respectively. In the MT-2-HIV-1ⅢB model, the CC50, IC50 and SI of Mango leaf extract were (174.13±22.36)μg/ml, (15.23±9.99)μg/ml and 11.44, respectively. Conclusion Mango leaf extract has low cytotoxicity in vitro and significant anti-HIV-1 activity, which has potential development and utilization value.
